摘要
为提高凝结芽孢杆菌固体发酵水平,通过单因素试验对发酵条件、培养基成分进行了筛选,再通过正交试验对培养基组成进行了优化。结果表明:固体发酵最佳条件为发酵温度35℃、培养基初始pH7.0、含水量40%、接种量30%和发酵时间48h,固体发酵后期(发酵36h时)选择40℃进行变温调控;最佳培养基组成为:培养基载体为麦麸(0.30kg),鱼粉2.00g/kg,磷酸氢二钾3.52g/kg,硝酸钠0.34g/kg,一水硫酸锰0.17g/kg。在此培养基组成和发酵条件下,凝结芽孢杆菌固体发酵菌体数达1.62×1010cfu/g。
To improve solid fermentation level of Bacillus coagulans, the fermentation conditions and culture medium compo- sition were screened by single factor methods, then the culture medium composition was optimized by orthogonal design. The re- sults showed that the optimal solid state fermentatiqn conditions were as: temperature 35℃, initial pH7.0, moisture content 40%, inoculation amount 30%, fermentation time 48 h, variable temperature control at 40℃ in the later fermentation period (at 36 h) ; the optimal culture medium composition as: wheat bran 0. 30 kg, fish meal 2. 00 g/kg, dipotassium phosphate 3.52 g/kg, sodium nitrate 0.34 g/kg, manganese sulfate monohydrate 0.17 g/kg. Under the optimized conditions,the of viable count could reach 1.62×10^10 cfu/g.
