摘要
目的:观察苦参碱对人胚肺成纤维细胞增殖和凋亡的影响。方法:选择苦参碱为干预药物,以不同浓度梯度作用于体外培养的人胚肺成纤维细胞,采用MTT法检测细胞增殖,用流式细胞仪分析细胞周期和凋亡率。结果:MTT结果显示,各干预组对HFL-1细胞生长均有显著抑制增殖作用,其强度呈时间-剂量依赖性。苦参碱作用于HFL-1细胞48 h后,各干预组细胞总凋亡率均明显高于对照组(P<0.05);与对照组比较,苦参碱各浓度组HFL-1细胞G0/G1期百分率显著上升(P<0.05)。结论:苦参碱可在一定程度上抑制人胚肺成纤维细胞增殖,并诱导其凋亡。
Objective:To observe the effect of matrine on prnliferation and apoptosis of human embryonic lung fibroblast. Methods:The human embryonic lung fibroblasts with matrine of different concentration were cultivated, cell prolifertion was measured by thiazolyl blue (MTT) assay;Cell cycle and apoptosis rate were determined with flow cytometre. Restults:MTT results showed that each intervention group has significantly inhibition effect on cell growth of HFL-I ,in dose-dependent and time-dependent manner. When treated with ma- trine of different concentration after 48 hours, the total apoptosis rate were significantly higher than control group( P 〈 0.05 ) ;compared with the control group,HFL-1 cells of marine concentration groups in G0/G1 phase percentage rise significantly( P 〈 0.05 ). Conclusion:Matrine can inhibit the proliferation of HLF-1 to a certain extent,and induce its apoptosis.
