HPLC法同时测定退黄丸中绿原酸、栀子苷的含量

Determination of Chlorogenic Acid and Geniposide in Tuihuang Pills by HPLC

目的:利用反相高效液相色谱法测定退黄丸中绿原酸、栀子苷的含量。方法:采用HPLC法,C18柱,B为甲醇,A为0.05%磷酸水溶液,梯度洗脱,在0～15 min,B相保持17%不变,15～30 min,B相从17%线性改变至25%,30～45 min,B相从25%线性改变至17%,45～55 min,B相保持17%10 min;柱温:25℃;流速:1.0 mL·min-1;检测波长:245 nm。结果:绿原酸、栀子苷线性范围分别为0.025 2～0.126 0 g·L-1、0.026 32～0.131 60 g·L-1;r值均为0.999 95,平均回收率为分别为98.80%、99.38%,RSD分别为1.18%、0.95%。结论:该方法操作简便,结果准确,重现性好,可用于退黄丸的质量控制。

Objective：To establish an RP-HPLC method for content determination of Chlorogenic acid and Geniposide in Tuihuang pills. Methods：The scopoletin was performed on a column C18 ;with a mobile phase of 0.05% H3 PQ - Methanol,in the first 15 min. Metha- nol keep 17% ,and in the next 15 min,where Methanol changed from 17% to 25% ,then in the next 10 min,where Methanol changed from 25% to 17% ,then keep Methanol 17% about 10 min;the column temperature：25 ℃ ;the flow rate was 1.0 mL · min -1 ;the detective wavelength was 245 nm. Results：The linear range of Chlorogenic acid and Geniposide was from 0. 025 2 -0. 126 0 g · L -1 and 0. 026 32 - 0.131 6 g · L-1 , r = 0. 999 95, and the average recovery was 98. 80% （ RSD = 1.18% ） ,99.38% （ RSD = 0.95% ）. Con- clusion：The method is accurate,fast and quantitative detection,which can be used in quality control of Tuihuang pills.
