煎煮时间对附子水煎液总生物碱和6种酯型生物碱含量的影响 被引量:6

摘要 目的:研究煎煮时间对附子水煎液总生物碱和6种酯型生物碱含量的影响。方法:采用酸碱滴定法测定总生物碱含量;采用HPLC,Agilent ZORBAX Extend-C18分析柱测定6种酯型生物碱含量。结果:随着煎煮时间的延长,不同炮制品中总生物碱的溶出率变化不明显;不同炮制品煎煮过程中酯型生物碱的变化规律不尽相同,其中乌头碱(AC)含量最低,水解最为彻底。结论:临床大剂量应用附子时,最大限度发挥其药理作用,恰如其分地掌握剂量与煎煮时间,是必不可少的。 Objective: To study the effect on total alkaloids and six kinds of ester - type alkaloids in water decoction of Aconitum carmichaeli Dexb. by different decocting time. Methods : The contents of total alkaloids were measured by acid base titration; the six kinds of ester - type alkaloids were tested by HPLC with Agilent ZORBAX Extend - C18. Results :As the extension of decoction time, the change of dissolution rate of total alkaloid in different processed drugs was not obvious. With the different processed drugs, the change law of ester type alkaloid in the process of the decoction was not the same ; the content of alkaloid was the lowest;the hydrolysis of alkaloid was most completely. Conclusion:It's essential to play maximum of pharmacological action and master dose and decoction time appropriately when apply of aconitum carmichaeli in clinic in a high - dose.
