紫癜颗粒对ITP小鼠外周血小板调控因子影响 被引量:4

摘要 目的:原发性血小板减少性紫癜(idiopathic thrombocytopenic purpura,ITP)是临床常见的出血性疾病,其发病机制尚未明确。该文从血小板计数和细胞因子考察ITP的发病机制以及紫癜颗粒对ITP小鼠的影响。方法:采用免疫造模法,建立慢性ITP模型。空白组、模型组按0.3 mL/20 g体重剂量胃饲蒸馏水,各给药组分别按12 g/kg体重剂量分别胃饲中药对照药、西药对照药和紫癜颗粒,每日1次,连续给药14 d。血细胞计数仪测定小鼠外周血血小板计数,双抗体夹心ELISA法检测骨髓白介素3(IL-3)、白介素6(IL-6)、白介素11(IL-11)、转化生长因子(TGF-β1)、血小板生成素(TPO)浓度。结果:各给药组药物均能提高ITP小鼠外周血血小板计数,有统计学意义(P<0.05);与模型组比较,各给药组血IL-3浓度明显降低,IL-6浓度明显降低,IL-11浓度明显降低,中药对照组和治疗组TGF-β1浓度明显降低,中药对照组和治疗组TPO浓度明显升高,有统计学意义(P<0.05);与各给药组比较,治疗组优于其他给药组,有统计学意义(P<0.05)。结论:中药对照药、西药对照药和紫癜颗粒均能够治疗ITP小鼠,以紫癜颗粒效果最优。紫癜颗粒能够升高ITP小鼠外周血小板计数,降低IL-3、IL-6、IL-11浓度和TGF-β1浓度,升高TPO浓度。 Objective : Idiopathic thrombocytopenic purpura is a common clinical hemorrhagic disease, and its pathogenesis is not clear. The platelet count and cytokines on the pathogenesis of ITP and purpura Grannie on ITP mice was studied. Methods : The mice anemia model was established by cyclophosphamide chemotherapy. The mice in normal group and model group were given distilled water by garage, and those in each treatment groups were given Danggui Buxue Decoction extracted by different methods for 14 days. Peripheral blood counting instrument was used to count blood cell. The ELISA was used to test concentrations of IL-2, EPO and TGF- β 1 of the bone marrow and the EPO concentration of the kidney. Results : Each group of drugs can improve the platelet count in peripheral blood ITP, there was statistical significance (P〈0.05). Compared with the model group, the treatment group's blood IL-3 concentrations were significantly decreased, IL-6 concentrations were significantly decreased, IL-11 concentration decreased significantly. The traditional Chinese medicine treatment group and the control group's TGF- beta l concentration was significantly reduced. Traditional Chinese medicine treatment group and the control group's TPO concentration increased significantly with statistical significance ( P〈0.05 ). And the treatment group was better than the other group, there was statistical significance ( P〈0.05 ) .Conclusion : Traditional Chinese medicine, western medicine controX drug and purpura granule can treat ITP mice with the optimal purpura granule effect. Purpura granule can improve ITP mice peripheral platelet count, can reduce the concentration of IL-3 and the IL-6 concentration, and decrease the concentration of IL-11, reduce the TGF- β 1 concentration, and increase the concentration of TPO.
