黄连素对胰腺癌细胞增殖及凋亡的影响 被引量:3

摘要 目的:探讨黄连素对胰腺癌细胞增殖及凋亡的影响。方法:用浓度分别为0μmol/L,25μmol/L,50μmol/L,100μmol/L,200μmol/L的黄连素与人胰腺癌Panc-1细胞共同培养,MTT法检测胰腺癌细胞的增殖抑制率,流式细胞仪检测细胞凋亡率,免疫组化检测细胞中的Caspase-3蛋白的表达。结果:黄连素对人胰腺癌Panc-1细胞增殖抑制率随药物浓度和作用时间的增加而增强;黄连素作用48 h后人胰腺癌Panc-1细胞凋亡率明显增加,且随药物浓度的增加而升高;黄连素作用48 h后人胰腺癌Panc-1细胞的Caspase-3蛋白表达增加,差异有统计学意义(P<0.05)。结论:黄连素能抑制胰腺癌Panc-1细胞的增殖,而且可能通过增加Caspase-3蛋白的表达来增强胰腺癌细胞的凋亡。 Objective To Investigate the effect of berberine on silence cell proliferation and apoptosis of pancreatic canc-er. Method pancreatic cancer cells were incubated with berberine at various concentration and time,the proliferation inhibition rate was as-sessed by MTT assay,apoptotic was analyzed by flow cytometry,Caspase -3 protein expressions was assessed by immunohistochemistry method. Results The proliferation of pancreatic cancer cells was significantly inhibited by berberine in a dose-dependent and time-de-pendent manner. A concentration-dependent increase of cell apoptosis was detected which was associated with a marked increment of the expression of Caspase-3 proteins,The difference was statistically significant(P〈0. 05). Conclusion Berberine can obviously inhibit the proliferation of pancreatic cancer cells. The activation and increase of Caspase-3 is the possible mechanism of apoptosis.
