摘要
目的探讨Wistar大鼠重症急性胰腺炎(SAP)肺损伤生存素(survivin)基因表达对血红素加氧酶1(HO-1)及细胞凋亡的影响。方法选择Wistar大鼠30只,随机分组后,建立SAP组24只(分为6h、12h、24h、48h,每组6只),对照组6只,利用逆转录-聚合酶链反应(RT-PCR)方法检测survivin基因的表达,免疫组织化学法检测HO-1的表达,DNA断裂的原位末端标记(TUNEL)法检测肺组织细胞凋亡的变化。结果 SAP各组survivin基因及HO-1表达量较对照组显著升高(P<0.05);细胞凋亡情况:对照组细胞结构正常,仅少数肺泡间质细胞及上皮细胞出现凋亡,SAP组细胞凋亡明显,且随时间推移逐渐增加。结论 SAP肺损伤中survivin基因显著升高,引起HO-1表达上升及细胞凋亡加重。提示SAP中survivin基因升高会引起肺损伤。
Objective To investigate the effects of survivin gene to the expression of heme oxygenase-1and cell apoptosis in severe acute pancreatitis(SAP)associated lung injury in Wistar rats.Methods The study enrolled 30 Wistar rats,after being randomized into groups,SAP group was established with 24 rats into four groups of 6h,12 h,24h,48h(6rats a group),and 6rats in control group.The expression of survivin gene was detected by RT-PCR,the expression of heme oxygenase-1 was tested with immunohistochemistry,TUNEL method was used to detect the apoptosis in lung tissue.Results Survivin gene and heme oxygenase-1expression in SAP group at different time were significantly higher than those of control group(P〈0.05).Cell apoptosis:the control group with normal structure of alveolar cells only a few part-time cells and epithelial cells apoptosis,apoptotic cells in SAP group were obvious,and increased with the time.Conclusion Survivin gene in SAP associated lung injury increased significantly,while the heme oxygenase-1expression increased,at the same time as the survivin gene expression level rises,the apoptotic cells increase.The data suggest that the increase of active survivin gene in SAP associated lung injury can cause lung injury.
