摘要
目的研究Rg1对IL-1β诱导的类风湿关节炎滑膜成纤维细胞(RASFs)表达细胞因子的调节作用。方法将RASFs分为空白对照(A)组、IL-1β20ng/ml对照(B)组、IL-1β20ng/ml+Rg1 0.4μg/ml(C)组、IL-1β20ng/ml+Rg1 4μg/ml(D)组、IL-1β20ng/ml+Rg1 40μg/ml(E)组,采用RT-PCR检测RASFs中TNF-α、IL-6、IL-8、IL-17、基质金属蛋白酶-3(MMP-3)和MMP-13的mRNA表达水平。结果与A组相比,B组TNF-α、IL-6、IL-8、IL-17、MMP-3和MMP-13mRNA水平升高(P<0.05),而Rg1呈浓度依赖性地抑制TNF-α、IL-6、IL-8和IL-17mRNA表达(P<0.05或P<0.01)。结论 Rg1抑制RASFs表达炎症因子,减轻关节炎症,也能抑制RASFs表达MMPs,改善骨侵蚀。
Objective To investigate the regulatory effect of Rgl on IL-1β-induced expressions of cytokines in synovial fibroblasts from the patients with rheumatoid arthritis(RASFS). Methods RASFs were divided into five groups of A(blank control),B(treated with IL-1β 20 ng/ml), C(treated with IL-1β 20 ng/ml plus Rg1 0. 4 μg/ml), D(treated with IL-1β 20 ng/ml plus Rg1 4 μg/ml) and E (treated with IL-1β 20 ng/ml plus Rg1 40 μg/ml). The mRNA expressions of TNF-α, IL-6, IL-8, IL-17, MMP-3 and MMP-13 in RASFs were detected by RT-PCR. Results Compared with group A, the mRNA expressions of TNF-α, IL-6, IL-8, IL-17, MMP-3 and MMP-13 were increased in group B (P〈0. 05),which were inhibited in a dose-dependent manner in groups of C, D and E(P〈0. 05 or P〈0. 01). Conclusion Rgl may improve the arthritis by inhibiting inflammatory cytokines and reduce the bone erosion via its suppression of MMPs expression in RASFs.
