摘要
通过靶向PML-RARα融合基因的小干扰RNA(siRNA)的体外干扰,研究其对急性早幼粒细胞白血病细胞NB4活性的影响。设计合成5对靶向PML-RARα融合基因的siRNA,转染NB4细胞,采用MTT比色法、软琼脂细胞集落形成实验检测siRNA对NB4细胞的增殖抑制作用,用流式细胞术测定细胞的凋亡,结果显示siRNA显著抑制了NB4细胞的增殖并有效诱导细胞凋亡。靶向PML-RARα融合基因的siRNA有望成为治疗急性早幼粒细胞白血病的有效方法。
This study aims to investigate the effects of small interference RNA (siRNA) targeting PML-RARa mRNA on the activity of the acute promyelocytic leukemia cell line NB4. The proliferation inhibition was evaluated by MTT assay and colony formation inhibiting test. Apoptosis was determined by flow cytometry after siRNA treat ment. The results showed that the cell growth of siRNA treated group was inhibited, and the apoptosis of NB4 could be induced. The siRNA targeting PML RARa mRNA might be a valid therapy of acute promyelocytic leukemia.
