线粒体功能损伤及靶向线粒体的抗氧化和抗癌药物研发进展 被引量:1

Research progress in mitochondrial damage and mitochondria-targeted anti-oxidant and anti-cancer therapeutic compounds

摘要 线粒体是一个十分重要但是目前被忽视的潜在治疗靶标,由于其基因组复制和组成方式较为独特,使线粒体容易受到生物毒性物质损害。线粒体功能损伤主要包括氧化性损伤,钙稳态失衡和ATP合成破坏。本文总结了引发线粒体功能失常的各种机制,线粒体功能紊乱与疾病的关系,论述了以线粒体为靶标的药物作用原理和效果;对靶向线粒体的抗癌、抗氧化药物存在的问题、应用前景等方面作了讨论。 Mitochondrion is an important but currently composition and replication style of mitochondrial genomes, substances. Mitochondrial damages include oxidative damage, neglected therapeutic target. Due to the unique mitochondria are vulnerable to biological toxic calcium dyshomeostasis and disruption to ATP synthesis. The underlying mechanisms which cause mitochondrial dysfunction, the relation between mitochondrial dysfunction and diverse pathologies as well as the strategies and effects of mitochondria-targeted compounds are reviewed in this paper. Lastly, the prospects and remaining problems of mitochondria-targeted anti-oxidant and anti-cancer compounds are discussed.
