摘要 目的:探讨Wnt5a及其受体Fzd2在环磷酰胺致大鼠神经管畸形神经上皮细胞中的表达变化及意义.方法:应用免疫荧光检测环磷酰胺处理对神经管畸形发生中Wnt5a及其受体Fzd2表达的影响.结果:与对照组相比,环磷酰胺处理后4、8h,神经管神经上皮细胞中Wnt5a和Fzd2阳性细胞光密度值无差异;处理后12和24 h,Wnt5a和Fzd2阳性细胞光密度值升高;处理后48 h,Wnt5a和Fzd2阳性细胞光密度降低.结论:Wnt5a及其受体Fzd2表达异常与环磷酰胺致大鼠神经管畸形密切相关. Objective:To explore the expression change and significance of Wnt5a and its receptor Fzd2 in the neuroepithelial cells of neural tube defects (NTDs) induced by cyclophosphamide in rats,and to provide new experimental evidence for the prevention and treatment of NTDs.Methods:The expression of Wnt5a and its receptor Fzd2 in the experimental group and the control group in the development of neural tube defects were detected by using immunofluorescence labeling technology.Results:Compared with the control group,at 4,8 hours after treatment,there was no significant change in the optical density of Wnt5a and Fzd2 positive cells in the neuroepithelial cells of the experimental group; at 12,24 hours after treatment,the optical density of Wnt5a and Fzd2 positive cells in the neuroepithelial cells of the experimental group increased,and the differences were all statistically significant; at 48 hours after treatment,the optical density of Wnt5a and Fzd2 positive cells in the neuroepithelial cells of the experimental group decreased,and the differences were statistically significant.Conclusion:The abnormal expressions of Wnt5a and its receptor Fzd2 are closely related to neural tube defects induced by cyclophosphamide.
