凋亡相关蛋白survivin和caspase-3在糖尿病大鼠下颌下腺的表达

摘要 目的:观察凋亡相关蛋白survivin和caspase-3在糖尿病大鼠下颌下腺的表达变化,探讨其与糖尿病发生的关系.方法:雄性SD大鼠腹腔内注射链脲佐菌素建立糖尿病模型,分别于注射后4周和12周测体质量和血糖,H-E染色观察下颌下腺的形态学变化,免疫组织化学SABC法检测survivin和caspase-3蛋白在下颌下腺的表达变化.结果:模型组大鼠下颌下腺组织明显萎缩,间质内纤维增生.在对照组下颌下腺导管上皮细胞高表达survivin和低表达caspase-3,模型组导管上皮细胞survivin的表达随病程延长显著减弱而caspase-3表达明显增强.结论:survivin和caspase-3在大鼠糖尿病模型下颌下腺的表达呈规律性变化,这与糖尿病的发病机制可能有着密切联系. Objective:To investigate the changes of apoptosis-ralated survivin and caspase-3 expression in the submandibular gland of diabetic rats,and explore the relationship to diabetes mellitus.Methods:A Sprague-Dawley diabetic rats model were induced by injection of streptozotocin.After 4 and 12 weeks,the rats were killed for assaying weight and blood sugar,and the submandibular glands were observed by H-E staining and the expression of survivin and caspase-3 by using immunocytochemistry methods.Results:In diabetic rats,submandibular gland atrophied and showed the significant proliferating interstitial fiber.Compared with the control group,survivin immunopositive cells in the submandibular gland showed a descending tendency with the progression of the disease,but caspase-3 showed an ascending tendency and immunopositive cells were increased more significantly in diabetes.Conclusion:survivin and caspase-3 expressions have a certain change in submandibular gland of diabetic rats,may be closely related to the pathogenesis of diabetes mellitus.
