摘要
目的:探讨过表达癌胚抗原相关黏附分子6(carcinoembryonic antigen-related cell adhesion molecule 6,CEACAM6)对胃癌细胞凋亡与失巢凋亡的影响。方法:构建CEACAM6真核过表达载体转染胃癌细胞SGC-7901和MKN-45,使其CEACAM6过表达,qRT-PCR、Western印迹验证过表达效果,免疫荧光定位CEACAM6;流式细胞仪检测过表达CEACAM6后胃癌细胞凋亡的变化,失巢凋亡检测其失巢凋亡的变化。结果:转染CEACAM6后,胃癌细胞SGC-7901和MKN-45的CEACAM6分别过表达了30 000倍和4 000倍;在胃癌细胞SGC-7901中凋亡与失巢凋亡分别下降了46.2%和53.8%,在MKN-45中凋亡与失巢凋亡分别下降了71.7%和74.6%。结论:过表达CEACAM6抑制胃癌细胞的凋亡和失巢凋亡,CEACAM6通过抑制胃癌细胞凋亡和失巢凋亡的途径参与胃癌细胞的恶性生长过程。
Objective To investigate the effects of overexpression of carcinoembryonic antigen-related cell adhesion molecule 6 (CEACAM6)on apoptosis and anoikis in gastric cancer (GC) cells. Methods CEACAM6 eukaryotic vector was constructed and transfected into SGC-7901 and MKN-45 GC ceils. The overexpression of CEACAM6 was confirmed by qRT-PCR and Western blot. The distribution of CEACAM6 was measured by immunofluorescence. Apoptosis and anoikis of GC cells were evaluated by flow cytometry (FCM) and CytoSelect^TM 24-Well anoikis assay respectively after overexpression of CEACAM6. Results CEACAM6 expression was successfully overexpressed by 30 000 folds in SGC- 7901 cells and 4 000 folds in MKN-45 cells, respectively. Overexpression of CEACAM6 inhibited apoptosis and anoikis by 46.2% and 53.8% in SGC-7901 GC cells and 71.7% and 74.6% in MKN-45 GC cells, respectively. Conclusions Overexpression of CEACAM6 inhibits apoptosis and anoikis of GC cells, which indicates that CEACAM6 participates in the malignant progression of GC cells through anoikis and apoptosis resistance.
