EB病毒相关肿瘤中EB病毒编码微小RNA的研究进展 被引量:3

摘要 EB病毒(Epstein—Barr virus,EBV)又称人类疱疹病毒4,是一种线性双链DNA病毒,在人群中广泛感染,与淋巴瘤、鼻咽癌、胃癌等多种恶性肿瘤的发生发展密切相关。微小RNA(microRNA,miRNA)是一类存在于多种真核生物和DNA病毒中的非编码RNA,长度为19—25nt,以碱基互补的方式和靶标mRNA的3‘端非翻译区(3’UTR)结合,导致靶标mRNA的翻译抑制或降解”。EBV是首个被发现可以编码miRNA的病毒。2013年6月miRbase数据库(www.mirbase.org)公布的数据显示,至今已经发现EBV编码的25个前体miRNA和44个成熟miRNA。EBVmiRNA主要由EBV基因组中的BHRF1和BART两个区域编码。BHRFl(Bam HI fragment H rightward open reading frame1)编码3个前体miRNA(ebv—miR—BHRF1-1,-2,-3)和4个成熟miRNA。BART(The Bam H I A region rightward transcript)编码两组包括22个前体miRNA和40个成熟miRNA:第1组位于BART外显子I和IB之间,编码8个前体miRNA(ebv-miR—BART1,-BART3-6,-BART15~17);第2组位于BART外显子IB和Ⅲ之间,编码13个前体miRNA(ebv—miR—BART7-14,-BART18-22)。ebv—miR—BART2则由BART外显子Ⅳ和V之间的区域编码。越来越多的证据表明,EBVmiRNA在调节EBV和宿主的基因表达并促进EBV相关肿瘤的发生发展方面发挥着重要的作用”。
