摘要
对大黄鱼精子超低温保存的抗冻液配方、冷冻程序和解冻程序进行了研究.试验结果表明:浓度为5%~20%的甘油都可以对大黄鱼精子的超低温冷冻保存起到保护作用.精子冷冻方法中,分步投氮法能获得较稳定的平均超过80%的激活率.不同解冻方法对精子的复苏率影响差异不显著.经3个月超低温冷冻保存的精子用于人工授精可以获得接近鲜精的授精率和孵化率.
The aim of the study was to study the key ingredients of antifreeze liquid,procedure of freezing and thawing for large yellow croaker sperm cryopreservation.The results demonstrated that diluents with different concentration of glycerol 5~20% can be used as cryoprotectants,and the postthaw sperm motility of frozen sperm with these cryoprotectants didn't show significant difference.By using the liquid nitrogen involved "three-step" method,the post-thaw sperm motility of frozen sperm is about 80%,which is similar to the of fresh sperm's.There is no significant difference between different tested thawing procedures.
