摘要
目的观察支气管哮喘(哮喘)大鼠肺组织中维甲酸受体相关孤儿受体(RORγt)和叉状头/翅膀状螺旋转录因子(Foxp3)的变化情况以及补肾平喘膏方对其的影响,以探讨补肾平喘膏方防治哮喘的作用机制。方法 60只SD大鼠随机分为正常组、模型组、西药组、膏方预防组、膏方治疗组和膏方防治(预防+治疗)组,每组10只。除正常对照组外,其余各组大鼠以卵蛋白(OVA)致敏并诱发哮喘模型。采用地塞米松或膏方治疗(正常组与模型组除外)后,RT-PCR法检测肺组织中RORγt和Foxp3的mRNA表达情况。结果模型组肺组织Foxp3 mRNA表达低于正常组(P<0.01),经膏方干预的各组大鼠肺组织Foxp3 mRNA表达升高(P<0.05),与正常组表达相似(P>0.05),膏方各组组间差异无统计学意义(P>0.05)。模型组肺组织RORγt mRNA表达高于正常组(P<0.01),经膏方干预的各组大鼠肺组织RORγt mRNA明显降低(P<0.05),膏方各组组间差异无统计学意义(P>0.05)。结论补肾平喘膏方可以通过调节哮喘大鼠RORγt/Foxp3平衡,改善Th17/Treg的失衡状态,从而防治哮喘。
Objective To observe the effects of medicinal extract on RORγt and Foxp3 in lung tissues of rats,and to explore its mechanism. Methods Sixty SD rats were randomly divided into normal group,model group,western medicine group,prevention group,treatment + prevention group,with ten rats in each group.Asthma was induced by intraperitoneal injection of ovalbumin(OVA) and forced inhalation of atomized OVA. Expression of RORγt and Foxp3 in lung tissues was detected by RT-PCR method. Results Expression of Foxp3 mRNA was lower in the model group was lower than that of the normal group( P〈0.01).Expressions of Foxp3 mRNA in medicinal extract groups were up-regulated(P〈0.05),without significant difference between normal group and medicinal extract groups(P〉0.05). Expression of RORγt mRNA was higher in the model group than that of the normal group(P〈0.01). Expressions of RORγt mRNA in medicinal extract groups were down-regulated(P〈0.05),and no significant difference was found among the medicinal extract groups(P〉0.05). Conclusion Medicinal extract for tonifying kidney to relieve asthma can prevent and treatment asthma by regulating the RORγt /Foxp3 balance and improving unbalance of Th17 / Treg.
