桑叶多糖对DN大鼠肾脏CTGF表达的影响 被引量:2

摘要 目的:观察桑叶多糖(mulberry leaves polysaccharide,MLP)对糖尿病肾病(diabetic nephropathy,DN)大鼠肾组织结缔组织生长因子(connective tissue growth factor,CTGF)基因表达的影响,探讨MLP抗肾纤维化的作用机制。方法:将Wistar大鼠随机分为正常对照组、模型组、MLP高、中、低剂量治疗组。采用一次性腹腔注射STZ的方法建立DN大鼠模型。给予不同剂量MLP进行治疗8w后,检测24h UAlb和尿蛋白含量;通过RT-PCR法测定肾脏中CTGF mRNA表达。结果:MLP能显著降低DN大鼠24h UAlb和尿蛋白含量(P<0.01),下调CTGF mRNA表达(P<0.01)。结论:MLP可减少DN大鼠尿蛋白、24h UAlb,降低肾脏CTGF mRNA的表达。 [Objective] To observe the effects of MLP on the expression of CTGF in the renal tissue of DN rats, and to investigate the antifibrosis mechanism of MLP. [Method] Wistar rats were randomly divided into the control, model, and high, middle, low doses of MLP groups. DN rat model was established by intraperitoneally injecting streptozotoein. After the DN rats were treated with different dose of MLP 8 weeks later, the change in urine protein was determined. The CTGF mRNA expression in the kidney were evaluated by RT-PCR. [Result] MLP could significantly reduce urine protein in DN rats ( P 〈 0. 01 ). MLP administration resulted in a remarkably down-regulation of CTGF mRNA expression ( P 〈0. 01 ) in the kidney of DN rats. [Conclusion] MLP could reduce urine protein and renal expression of CTGF mRNA in rats with DN.
