回药烫伤膏中大黄与黄芩提取工艺研究 被引量:2

摘要 研究了回药烫伤膏中大黄与黄芩的提取工艺。采用乙醇回流法对大黄与黄芩进行合并提取,通过单因素和L9(34)正交试验考察了乙醇浓度、乙醇用量、提取时间和提取次数对提取工艺的影响。应用紫外分光光度法测定烫伤膏药材乙醇提取液中大黄素和黄芩苷的含量,并对该含量测定方法进行了考察。通过对乙醇提取液进行浓缩干燥处理计算干膏率。以大黄素与黄芩苷含量及干膏率为考察指标,结合多指标加权平分法(提取液中大黄素含量∶提取液中黄芩苷含量∶干膏率的权重系数为0.3∶0.3∶0.4)优化大黄与黄芩的提取工艺。优化得到的提取工艺为用大黄与黄芩饮片总量8倍量的600mL/L乙醇回流提取2次,每次提取1.5h。验证试验表明该工艺稳定、合理可行,可用于回药烫伤膏生产。 To study the extraction process of rhubarb and radix scutellariae in hui medicine scald ointment, the ethanol refluxing was used to extract Rhubarb and Scutellariae,through the single factor and L9 (3 4 ) orthogonal test to evaluate the effects of alcohol concentration,alcohol amount,extraction time and ex-traction times on extraction process.The UV spectrophotometric method was used to determine the content of emodin and baicalin in scald ointment extraction solution,and the method for determining the content was investigated.The dry extract rate was calculated through the ethanol extract solution which is in the process of concentrating and drying.The emodin and baicalin contents and dry paste rates was used as ex-amining indexes,we combined with the multi-index weighted bisection method (emodin content in extract solution:baicalin content in extract solution:dry weight coefficient of paste rate is 0.3∶0.3∶0.4)to op-timize the extraction process of rhubarb and scutellariae.After optimizing,the extraction process was as followings.Rhubarb and Scutellaria crude slices total 8 times of 60% ethanol reflux extraction 2 times, each time 1.5 h.Verification test showed that the process is stable,reasonable and feasible,and can be used for medicine scald ointment production.
