摘要
通过观察川芎嗪对膝关节骨性关节炎(OA)模型大鼠关节液中一氧化氮(NO)和前列腺素E2(PGE2)的影响,探讨川芎嗪治疗OA的作用机制。将24只SPF级健康雌性SD大鼠随机分为假手术组、OA模型空白组和川芎嗪组。通过切除双侧卵巢并切断右侧膝内侧副韧带及交叉韧带建立膝关节骨性关节炎动物模型,川芎嗪组于术后第4周开始关节腔注射川芎嗪,OA模型空白组与假手术组注射等量的生理盐水,分别于术后第4、8周抽取关节液作NO、PGE2的测定,对比各组数据。术后8周,关节液中NO、PGE2的水平在川芎嗪组中表达均较OA模型空白组及术后第4周川芎嗪组明显减弱(P<0.05,P<0.01);OA模型空白组及川芎嗪组关节液中的NO、PGE2均较假手术组明显升高(P<0.01);术后8周川芎嗪组关节液中的NO、PGE2与假手术组比较均无统计学差异。川芎嗪能明显降低骨关节炎关节液中NO、PGE2的含量水平,对减轻OA炎症有一定的作用。
2.The Affiliated Hospital of Orthopedics and Traumatology ,Guangzhou University of Chinese Medicine ,Guangzhou,Guangdong,510240,China) Through observing the effect of ligustrazine on the level of nitric oxide (NO)and prostaglandin E2(PGE2)in synovial fluid of knee osteoarthritis model in rats,to investigate the mechanism of ligus-trazine in the treatment of osteoarthritis (OA),twenty-four SD rats were randomly divided into sham oper-ation group,OA model blank group and ligustrazine group.By ovariectomy and cutting off the right knee of tibial collatera lligament and cruciate ligament,the model of osteoarthritis of knee was made.The ligus-trazine group were treated with ligustrazine injection,the sham operation and the OA model blank group were treated with the same volume of saline in 4 weeks after operation were taken.Synovial fluid extrac-tions at 4 and 8 weeks after operation,for the determination of NO and PGE2,which data were used to contrast.The synovial fluid NO and PGE2 levels of expression in the ligustrazine group in the 8 weeks af-ter operation decreased significantly compared with OA model blank group and the after 4 weeks of ligus-trazine group(P 〈0.05,P 〈0.01).The synovial fluid NO,PGE2 levels of expression in the OA model blank group and ligustrazine group were significantly higher than those in the sham operation group(P 〈0.01).The synovial fluid NO,PGE2 levels of expression in the ligustrazine group in the 8 weeks after op-eration compared with the sham operation group had no statistical difference.Ligustrazine can significantly decrease NO,PGE2 contents in the synovial fluid of OA,and has certain effect to reduce the OA inflam-mation.
