心肌梗死后小鼠心肌Bax和Bcl-2的表达 被引量:1

Expression of Bax and Bcl-2 in mice with myocardial infarction

摘要 目的研究心肌梗死后小鼠心脏中Bax和Bcl-2的表达。方法雄性C57BL/6小鼠40只随机分为2组:对照组(10只)和实验组(30只),实验组结扎冠状动脉左前降支建立心肌梗死模型,对照组仅在冠状动脉前降支处穿过缝线,不做结扎。术后4周对2组小鼠进行心肌取材,Western blot方法检测Bax和Bcl-2蛋白在心脏表达的差异。结果实验组小鼠术后4周存活率为53.3%(16/30),对照组为100%(10/10)。Bax和Bcl-2在对照组心肌均有表达,在实验组小鼠心脏远离梗死区(N)、心肌梗死周边区(B)、梗死区(I)的表达逐渐增加(P<0.05),且B区、I区Bax/Bcl-2比值高于N区及对照组(P<0.05)。结论 Bax和Bcl-2在心肌梗死后心脏中呈高表达,且Bax/Bcl-2比值在心肌梗死区及周边区显著增加,提示凋亡可能在心肌梗死后心脏重构中发挥重要作用。 Objective To determine the expression of apoptotic protein Bax and Bcl-2 in heart of mice after myocar-dial infarction.Methods Forty male C57BL/6 mice were randomly divided into 2 groups: the control group (10 rats) and experimental group (30 rats), in the experimental group, left anterior descending coronary artery was ligated to establish the model of myocardial infarction , in the control group , no ligation were performed in the anterior descending coronary artery .Af-ter 4 weeks, the 2 groups of mice&#39;s myocardial material were taken , western blot method were used for detection of Bax and Bcl-2 protein expression differences in the heart .Results 4 weeks after operation , the survival rate of mice in the experimen-tal group was 53.3%(16/30), the control group was 100% (10/10).Bax and Bcl-2 in the control group had the expres-sion,in the experimental group ,expression increased gradually in area far from myocardial infarcted ( N) ,the surrounding area of myocardial infarction (B), infarction area (I) ( P 〈0.05), and the B zone, I zone Bax/Bcl-2 ratio is higher than that of N and control group ( P 〈0.05).Conclusion The high expression of Bax and Bcl-2 in heart after myocardial infarction may suggest that apoptosis plays an important role in later period of cardiac reconstruction after myocardial infarction .
