二至百合浓缩膏对雌性去势大鼠性激素与下丘脑神经递质的调节作用

摘要 目的:探讨二至百合浓缩膏对雌性去势大鼠(双侧卵巢切除术)子宫、性激素水平及下丘脑神经递质的影响和作用机制。方法:3~4月龄Wistar雌性大鼠随机分成6组,假手术组,模型组,更年欣胶囊组,二至百合高剂量组、中剂量组、低剂量组。除假手术组,其余各组手术摘除双侧卵巢建立去势更年期大鼠模型,分别每天灌胃相应受试样品,连续45 d。摘除各组大鼠子宫称质量并计算子宫系数,测定各组大鼠血清雌二醇(E2)、促卵泡生成激素(FSH)、促黄体生成激素(LH)水平和动物下丘脑中去甲肾上腺素(NE)、多巴胺(DA)和5羟色胺(5-HT)的含量。结果:与假手术组比较,二至百合3个剂量组均可不同程度升高血清E2水平(P<0.05),并且均可显著降低LH水平(P<0.01);二至百合高剂量组、中剂量组均可降低血清FSH水平(P<0.01)。与模型组比较,二至百合高剂量组可显著升高大鼠子宫脏器系数(P<0.05),同时能不同程度降低下丘脑组织中NE、DA、和5-HT的含量(P<0.05)。结论:二至百合浓缩膏能明显纠正下丘脑单胺类神经递质的紊乱,对模型大鼠生殖内分泌紊乱具有较好地调节作用。 Objective : To study the effect and action of Erzhibaihe concentrated paste on uterus, reproductive endocrine and hypothalamic of female castrated rats. Methods:3 -4 months old female wistar rats were randomly divided into six groups with shamoperated group, model group, gengnianxin capsule group, Erzhibaihe high dose, middle dose and low dose. All rats were established the models of climacteric syndrome with ovariectomized surgical except sham-operated group. They were given test sample for 45 days. Then weighing the uterus of rats which were excised and calculating the uterus coefficients, estradiol ( E2 ), follicle-stimulating hormone ( FSH), luteinizing hormone (LH) levels of serum and hypothalamic norepinephrine in elements ( NE), dopamine (DA) and serotonin ( 5- HT) levels were determined. Results:Compared with sham-operated group,three dose groups of erzhibaihe can increase serum E2 levels ( P 〈 0. 05 ) , and reduce LH levels significantly ( P 〈 0. 01 ) ; Erzhibaihe concentrated paste high dose and middle groups both can reduce serum FSH levels (P 〈 0.01 ). Compared with model group, Erzhibaihe concentrated paste high dose group can increase rat uterine organ coefficient ( P 〈 0. 05 ), while can reduce the hypothalamus tissue NE, DA and 5-HT levels ( P 〈 0.05 ). Conclusion : Erzhibaihe concentrated paste can obviously rectify the disorder of hypothalamic neuro modulator, and regulate the reproductive endocrine disorders of model rats.
