摘要
目的 探讨沉默信息调节因子1 (silent information regulator 1,sirt 1)与抗氧化酶在肾脏衰老中的作用.方法 选择SD雄性大鼠20只,将10只大鼠饲养至3月龄(相当于成人20岁)作为青年组,10只大鼠饲养至24月龄(相当于成人80岁以上)作为衰老组.利用肾脏组织匀浆测定还原型谷胱甘肽(gluathione,GSH)含量,Western Blot检测肾脏组织谷胱甘肽过氧化物酶(GPx 1)、GCLC、谷氨酸半胱氨酸连接酶(GCLM)、醌类氧化还原酶(NQO 1)和Sirt 1蛋白质表达.结果 肾脏组织GSH含量测定结果显示:3月龄青年组大鼠肾脏组织GSH含量为(23.17±1.61) μg/mg,24月龄衰老组大鼠肾脏组织GSH含量为(16.39±1.39) μg/mg,衰老组肾脏组织GSH含量明显降低,2组比较有统计学差异(P-0.029);同时衰老组肾脏GPx 1、GCLC、GCLM和Sirt 1蛋白质表达均较对照明显降低(P值分别为0.023、0.001、<0.001和0.002).而NQO1蛋白表达较对照组显著增加(P-0.001).结论 肾脏衰老的发生可能与沉默信息调节因子1及抗氧化酶表达减少有关.
Objective To observe the expression of silent information regulator 1 (Sirt1) and antioxi dant enzymes in the kidney aging.Methods Rat aging model was established using 3-,or 24-month old rats as young group and aging group respectively (n=10 each).The amount of GSH in kidney homogenate was tested.Western blotting was used to detect the protein expression of GPx1,GCLC,GCLM,NQO1 and Sirt1.Results Renal GSH contents in aging group were significantly lower than those in young group [(16.39± 1.39) μg/mg vs.(23.17 ± 1.61) μg/mg,P-0.029].Western blotting revealed that the ex pression levels of GPx1,GCLC,GCLM and Sirt1 proteins in the kidney of aging group was decreased appar ently (P =0.023,0.001,<0.001,and 0.002 respectively),and NQO1 protein level was significantly in creased (P=0.001) as compared with those in young group.Conclusions Decreased expression of Sirt1 and antioxidant enzymes may play a role in the kidney aging.
