摘要 β-葡糖醛酸酶(GUS)报告系统是现代分子生物学研究领域中被广泛使用的一种重要工具,在解析基因时空表达调控的研究中发挥着重要作用.本文概述了报告基因GUS的生化特性及检测手段,从启动子元件鉴定、基因诱捕、无标记转基因技术等方面论述GUS的应用现状和优势,并针对内源GUS、GUS抑制因子等问题和改进优化手段进行了分析,为该技术在植物功能基因研究中进一步拓展提供新线索和思路. Beta-glucuronidase(GUS) reporter system is widely used in modern molecular biology, and playing vital roles in analysis of regulation of gene expression in time and space. This paper analyzed and summarized detection, biochemical characteristics, application and advantages of GUS from promoter element identification, gene trap and marker free transgenic technology. Meanwhile, some problems such as endogenous GUS and GUS inhibitors were discussed, and the optimization methods were also included. This review will provide new clues and ideas for further research on functional gene in plants.
