摘要
目的:观察藏药波棱瓜子乙酸乙酯提取物对卡介苗(BCG)加脂多糖(LPS)联合诱导免疫性肝损伤小鼠的保护作用。方法:采用卡介苗联合脂多糖诱导造成免疫性肝损伤小鼠模型,ig给予波棱瓜子乙酸乙酯提取物(0.25、0.35、0.51g/kg)连续12d,比较各组脏器指数,测定小鼠血清中丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、一氧化氮(NO)的含量及肝脏组织匀浆中超氧化物歧化酶(SOD)、丙二醛(MDA)含量,HE染色切片观察肝组织病理学变化。结果:波棱瓜子乙酸乙酯提取物(0.25、0.35、0.51g/kg)可显著降低免疫性肝损伤小鼠的肝脏指数及脾脏指数;改善肝脏组织病理学变化;降低肝损伤小鼠血清中ALT、AST水平;升高肝匀浆中SOD活性,降低MDA含量;但对血清中NO含量无显著影响。结论:波棱瓜子乙酸乙酯提取物对BCG+LPS诱导小鼠免疫性肝损伤有明显的保护作用。
Objective: To investigate the protective effects of Herpetospermum ethyl acetate extracts on immunologic liver injury model. Methods:The model of mouse immunological liver injury were induced by injection of bacillus calmette guerin( BCG) and lipopolysaccharide( LPS)from tail vein. The organ index of mice were detected. The alanine aminotransferase( ALT),aspartate aminotransferase( AST),superoxide dismutase( SOD),malondialdehyde( MDA),nitrogen monoxidum( NO) content were assayed by spectrophotometry,hepatic tissues stained with hematoxylin and eosin( HE) were prepared for histological analysis to liver injury. Results: HE( 0. 25,0. 35,0. 51g /kg) could decrease liver index,spleen index and the ALT,AST,MDA content,prevented the reduction of SOD activities. Meanwhile,the pathological of hepatic tissues were revesed. But HE has no effect on the contents of NO in the blood serum. Conclusion: HE showed invariably protective action on immunological liver injury in mice.
