维吾尔族妇女子宫颈病变中CTGF、IL-1β、Agr和Adi-poR1表达与HPV16感染的相关性 被引量:5

Expression of CTGF,IL-1β,aggrecan and adiponectin receptor 1 in cervical lesions and their association with HPV16 infection in Uyghur women

摘要 目的探讨结缔组织生长因子(connective tissue growth factor,CTGF)、白细胞介素-1β(interleukin-1β,IL-1β)、蛋白聚糖聚合物(aggrecan,Agr)和脂联素受体1(adiponectin receptor 1,AdipoR1)在维吾尔族妇女子宫颈病变中的表达与HPV16感染、子宫颈癌分化程度的关系。方法采用免疫组化SP法检测CTGF、IL-1β、Agr和AdipoR1在喀什维吾尔族17例子宫颈炎和46例子宫颈癌组织中的表达;采用PCR技术检测相应标本中HPV16的感染。利用图像分析软件IPP 6.0半定量检测子宫颈病变中CTGF、IL-1β、Agr和AdipoR1的累积光密度(IOD SUM)值。结果 PCR检测结果显示子宫颈癌中HPV16阳性者22例,阴性者24例;子宫颈炎中HPV16阳性者5例,阴性者12例。CTGF在子宫颈癌组中的表达低于子宫颈炎组(P<0.01)。IL-1β、Agr在子宫颈癌组的表达高于子宫颈炎组(P<0.01)。CTGF在HPV16阳性子宫颈癌组的表达高于HPV16阴性组(P<0.05)。AdipoR1在HPV16阳性子宫颈炎组的表达高于HPV16阴性组(P<0.05)。CTGF、IL-1β、Agr和AdipoR1在子宫颈癌中的表达与分化程度无相关性(P>0.05)。结论与子宫颈炎组织相比,子宫颈癌中CTGF表达量下降,IL-1β和Agr表达量上升,在子宫颈癌的发生、发展中起一定的作用,其检测结果对维吾尔族妇女子宫颈癌诊断有一定的参考价值。 Purpose To study the expression levels of connective tissue growth factor (CTGF), IL-1β, aggrecan (Agr) and AdipoR1 in cervical cancer tissues and their correlation with HPV16 infection in Uyghur women. Methods We performed semi-quantitative studies of expression of CTGF, IL-1β, Agr and AdipoR1 in paraffin-embedded cervical tissues obtained from 17 cervicitis and 46 cervical cancer patients using immunohistochemical S-P staining and image analysis by Image-Pro Plus 6. 0 software to determine the sum integrated optical density (IOD). All the study subjects were Uyghur patients from Kashghar region of Xinjiang. The Infection of HPV was determined by examination of HPV DNA in cervical samples with PCR using HPV specific primer set. Results Analysis of HPV16 infection showed that there were 22 cases were positive and 24 were negative for HPV16 infection in cervical cancer patients. Whereas in patients with cervicitis, only 5 cases were positive and 12 were negative for HPV16 infection. Expression of CTGF in cervical cancer tissues was lower than that in cervicitis tissues ( P 〈 0. 01 ). The expression of IL-1β and Agr in cervical cancer tissues was higher than that in cervicitis tissues (P 〈 0. 01 ). Expression of CTGF in cervical cancer patients with HPV16 infection was higher than that in cervical cancer patients with HPV negative (P 〈 0. 05 ). Expression of AdipoR1 in HPV16 positive patients with cervicitis was higher than that in cervicitis patients with HPV16 negative (P 〈 0. 05). There was no obvious correlation between the expression of CT- GF, IL-1β, Agr and AdipoR1 in cervical cancer patients with the differentiation degree (P 〉 0. 05). Conclusion Compared to patients with cervicitis, expression of CTGF in cervical cancer was down-regulated, while the expression of IL-1β and Agr was up-regulated, implying that these molecules may play certain roles in the occurrence and development of cervical cancer, and their detection may have certain levels of diagnostic values for cervical cancer in Uyghur women.
