Correlation of MDM2 and epithelial-mesenchymal transition-related biomarkers in non small cell lung cancer

摘要 目的探讨非小细胞肺癌(NSCLC)组织中上皮间质转化(EMT)相关分子标记物和鼠双微体基因2(MDM2)蛋白的表达及其临床意义。方法采用免疫组化法检测80例NSCLC及20例癌旁正常肺组织中E-钙黏蛋白、N-钙黏蛋白、波形蛋白和MDM2蛋白的表达,分析MDM2表达与EMT相关分子标记物表达的相关性。结果 NSCLC组织中E-钙黏蛋白、N-钙黏蛋白、波形蛋白和MDM2蛋白阳性表达率分别为57.5%、81.3%、38.8%和71.3%,均低于癌旁组织中的76.3%、68.8%、21.3%和53.8%(P<0.05)。在NSCLC组织中,N-钙黏蛋白和波形蛋白表达与MDM2蛋白表达均呈正相关(r=0.405,r=0.331,P<0.05),而E-钙黏蛋白表达与MDM2蛋白表达呈负相关(r=-0.339,P<0.05)。结论在NSCLC中,MDM2过表达可能促进了肿瘤的EMT。联合检测MDM2和EMT相关分子标记物,可以作为判断NSCLC的恶性程度、转移潜能以及预后的重要参考指标。 Objective To study the expression of epithelial-mesenchymal transition(EMT)-related biomarkers and murine double minute 2(MDM2)in non-small cell lung cancer(NSCLC)and its clinical significance.Methods The protein expressions of E-cadherin,N-cadherin,vimentin and MDM2 were detected by immunohistochemistry in 80 samples of NSCLC and 20 samples of canceradjacent normal tissues.The correlation between MDM2 expression and EMT-related biomarkers was analyzed.Results The positive expression rates of E-cadherin,N-cadherin,vimentin and MDM2 in NSCLC tissues were 57.5%,81.3%,38.8%and 71.3%,which were significantly lower than 76.3%,68.8%,21.3%and 53.8%(P〈0.05).In NSCLC,the expressions of N-cadherin and vimentin were positively correlated to that of MDM2(r=0.405,r=0.331,P〈0.05),while E-protein expression was negatively correlated with MDM2(r=-0.339,P〈0.05).Conclusion In NSCLC,the overexpression of MDM2 may promote tumor EMT.The combined detection of MDM2 and EMT-related molecular markers may be taken as an important reference in evaluating the malignancy,metastasis potentiality and prognosis of patients with NSCLC.
