摘要
目的探讨美满霉素(MC)对脂多糖(LPS)诱导的BV-2小胶质细胞肿瘤坏死因子-α(TNF-α)表达的影响。方法采用相应浓度的MC或LPS(100 ng/ml)对MC组及0.1、1、10、100μmol/L MC+LPS组、LPS组、空白对照组BV-2小胶质细胞进行预处理。ELISA法检测BV-2小胶质细胞TNF-α蛋白的表达,逆转录-PCR检测细胞TNF-αmRNA表达。结果与空白对照组比较,0.1、1μmol/L MC+LPS组及LPS组的TNF-α水平显著升高(均P<0.01)。10、100μmol/L MC+LPS组TNF-α水平显著低于LPS组(均P<0.01)。与空白对照组比较,LPS组及10μmol/L MC+LPS组TNF-αmRNA的表达水平显著增高(均P<0.01)。10μmol/L MC+LPS组TNF-αmRNA表达水平显著低于LPS组(P<0.01)。结论 MC预处理(≥10μmol/L)可显著抑制LPS诱导的小胶质细胞TNF-α的表达。
Objective To explore the effect of minocycline (MC) on lipopolysaccharide (LPS)-induced expression of tumor necrosis factor (TNF)-α
in BV-2 mieroglial cells. Methods The BV-2 microglial cell were pretreatment with corresponding concentration MC or LPS (100 ng/ml) in MC group and 0. 1, 1, 10, 100 μmol/L MC + LPS group, LPS group and blank control group. The expression of TNF-α in BV-2 mieroglial cell was detected by ELISA, and the expression of TNF-α mRNA was detected by reverse transcription ( RT )-PCR. Results Compared with blank control group, the expression of TNF--α in 0. 1, 1 p, mol/L MC + LPS group and LPS group were significantly increased ( all P 〈 0. 01 ). The expression of TNF-α in 10, 100 μmol/L MC + LPS group were significantly decreased than that in LPS group ( all P 〈 0. 01 ). Compared with blank control group, the expression of TNF-α mRNA in 10 μmol/L MC + LPS group and LPS group were significantly increased ( all P 〈 0. 01 ). The expression of TNF-α mRNA in 10 μmol/L MC + LPS group were significantly decreased than that in LPS group ( P 〈 0. 01 ). Conclusion The MC pretreatment ( ≥10 μmol/L ) can significantly inhibit the the expression of TNF-oL with LPS-induced production of TNF-α in microglial cell.
