特异性阻抑DKK-1在激素性股骨头坏死治疗中的应用与展望 被引量:2

Application and prospects of specific inhibition of DKK-1 in the treatment of glucocorticoids-induced osteoneerosis of the femoral head

摘要 Wnt信号通路对成骨细胞的发育及分化起着重要作用。作为Wnt信号通路的抑制因子DKK-1能抑制骨形成,导致骨质疏松,而通过拮抗DKK-1的作用可使骨量增加。近年来的研究发现,糖皮质激素(GCs)诱发股骨头坏死(ONFH)与GCs上调胞外蛋白DKK-1表达密切相关。特异性阻抑DKK-1基因表达可能成为治疗GCs诱发的ONFH的新靶点。本文就特异性阻抑DKK-1基因表达对Wnt信号通路的调控及在激素性股骨头坏死治疗中的应用及展望做一综述。 The Wnt signaling pathway plays a vital role in the process of osteoblast proliferation and differentiation,this pathway can be impeded by an inhibitor,DKK-1. Over-expression of DKK-1 could affect bone formation and even lead to osteopenia. In recent years,a number of studies have found that glucocorticoids( GCs) up regulation to the extracellular protein DKK-1 expression was linked to the occurrence of GCs-induced osteoneerosis of the femoral head( ONFH). The expression of specific inhibition of DKK-1 gene may be a new target for the treatment of GCs-induced ONFH. Here,we reviewed the specific inhibition of DKK-1 gene expression in regulation of Wnt signaling pathway and application and prospects of specific inhibition of DKK-1 in the treatment of GCs-induced ONFH.
