摘要
目的探讨氟伐他汀对大鼠角膜碱烧伤后Smad-3表达的影响。方法 24只SD大鼠随机分为3组:生理盐水组、氟伐他汀组和地塞米松组,每组8只,用1mol/L的NaOH烧伤大鼠右眼,生理盐水组、氟伐他汀组和地塞米松组分别以生理盐水、氟伐他汀和1g/L地塞米松点眼。烧伤后1~7 d用裂隙灯观察角膜新生血管的生长情况,HE染色观察角膜组织病理学改变,免疫组化观察Smad-3的表达情况。结果烧伤后第7天,氟伐他汀组的角膜新生血管生长密度显著低于生理盐水组,差异有统计学意义,P〈0.05。组织病理学检查结果提示,氟伐他汀组与生理盐水组相比,新生血管的形成明显减少,管腔小且稀疏,周围炎症细胞浸润明显减少,角膜组织结构明显整齐。免疫组化结果显示氟伐他汀组的Smad-3的表达情况显著降低。结论氟伐他汀能通过下调Smad-3的表达,来减轻大鼠角膜碱烧伤后新生血管的生成。
Objective To investigate the effects of fluvastatin on expression of Smad-3 in alkali-burned cornea in rats. Methods Alkali corneal burns were induced to right eye in 24 SD rats using 1 mol /L NaOH. Animals were then randomly received one of 3 treatments( n = 8 for each group) : normal saline,fluvastatin,or dexamethasone. In 1 ~ 7days,corneal neovascularization was examined with slit-lamp,and pathological changes were evaluated with HE staining.The expression of Smad-3 was detected with immunohistochemical staining. Results At 7 days after corneal burns,corneal neovascularization was significantly less severe in rats treated with fluvastatin than in rats treated with normal saline( P〈 0. 05). Histopathological examines showed less neovascularization and inflammatory cells infiltration,and more orderly corneal structure. Immunohistochemical staining showed that the expression of Smad-3 was significantly lower in fluvastatin treated rats. Conclusion Fluvastatin can reduce the severity of corneal neovascularization,and the mechanism may be related to the down-regulated expression of Smad-3.
