球形孢子丝菌64株由菌丝相向酵母相的转化实验 被引量:1

摘要 目的观察球形孢子丝菌由菌丝相向酵母相转换形态学的变化,在三种培养基的酵母转化率。方法将64株球形孢子丝菌于脑心浸液琼脂(BHI)35℃观察菌落及显微镜下酵母转化形态,并与0.5%葡萄糖脑心浸液琼脂(GSBHI)、马龄薯(PDA)培养基上酵母相转化率进行比较。结果多次传代能使酵母转化更完全,BHI转化率48.43%,GSBHI转化率65.63%,PDA转化率为20.31%。结论球形孢子丝菌由菌丝相至酵母相转化需经过连续多次传代;在GSBHI培养基中35℃酵母转化率最高。 Objective To observe the morphology transformation from mycelia phase to yeast phase of s. globosa and the conversion rate of it in three kinds of culture medium. Methods Total 64-strains of s. globosa were inoculated in the Brain Heart Infusion Agar( BHI ) in 35℃ and colony of yeast was observed under-microscoped. The result of conversion rate of the yeast was compared with that inoculated with(0. 5% GSBHI) / (PDA). Results High passage can make the yeast conversion more completely. The conversion rate of BHI was 48.43%, GSBHI was 65.63% and PDA was 20. 31%. Conclusion Multiple successive are required to make s. globosa conversion from mycelial phase to yeast phase; and the conversion rate is highest with GSBHI culture medium in 35℃.
