摘要
目的探讨缺氧诱导因子1α(HIF-1α)和Survivin表达活性与喉鳞癌浸润及转移行为的关系。方法 86例喉鳞癌及32例正常喉黏膜上皮组织标本,应用免疫组化染色技术检测HIF1α和Survivin表达活性,分析其与患者临床病理特征的关系。结果喉鳞癌组织中,HIF1α与Survivin阳性表达率均明显高于正常喉黏膜上皮组织(P<0.05),且其阳性表达率与肿瘤临床分期、淋巴结转移明显关(P<0.05),而Survivin阳性表达率更与肿瘤病理分级显著相关(P<0.05)。HIF1α和Survivin表达水平呈正相关(r=0.456,P<0.05)。结论喉鳞癌组织HIF-1α活性升高,可能通过诱导Survivin高表达而对喉鳞癌细胞的侵袭、转移生物学行为发挥促进作用。
Objective To investigate the association of expressive activities of hypoxia-inducible factor 1α (HIF-1α) and survivin in the tissue of human laryngeal squamous cell carcinoma (LSCC) with clinical pathological features of tumor in such biological behaviors as invasion and metastasis. Methods Included in this study were 86 cases of LSCC and 32 cases of healthy laryngeal epithelial tissue samples, with the expressive activities of HIF-1α and survivin determined by immunohistochemical procedures. Then, the possible association of them with the behavior features of tumor in invasion and metastasis was evaluated carefully in the term of clinical pathology. Results The positive expression rates of HIF-1α and survivin was both significantly higher in the tissue samples of laryngeal squamous cell carcinoma than that in normal laryngeal epithelial tissue samples (P〈0.05), with significant correlation of positive expression rate of them with clinical TNM staging and neck lymph node metastasis (P〈0.05). Moreover, the positive expression rate of survivin was obviously correlated with histological grade of tumor cells further (P〈0.05). There was a positive correlation of the expressive level of HIF-1α with that of survivin (r=0.456, P〈0.01). Conclusions The expressive activity of HIF-1α is significantly increased in the tissue of LSCC. Then, expressively elevated HIF-1α may promote the expression of malignant behavior of tumor cells, especially in local invasion and distal metastasis, through inducing up-regulated survivin expression.
