摘要
为了解磷酸烯醇式丙酮酸羧激酶1(PCK1)基因在冷刺激下肉鸡组织中的表达情况,选择75只AA肉鸡随机分成5组,其中1组为对照组(正常饲养温度),其余4组为处理组(比正常饲养温度低3℃),于8日龄起每天分别进行冷刺激1h、3h、5h和24h,21日龄结束,22日龄屠宰测定PCK1基因在各组织的表达情况。结果表明:在冷刺激条件下,PCK1基因表达量存在组织特异性,在肝脏中表达量最高,其次是胸腺,第三是肾脏。在相同组织中,除冷刺激24h处理组外,心脏中PCK1基因的表达量随冷刺激时间的延长变化幅度不大,肝脏、肾脏和肺脏中呈上升趋势,脾脏中呈下降趋势,胸腺中呈先下降后上升趋势。对于参与糖异生的肝脏和肾脏,冷刺激对其中PCK1基因的表达量有显著上调作用。
75 AA broilers were divided into five groups (one group as CK and four groups as test groups)randomly and averagely,and the expression of PCK 1 gene of different AA broiler's organs under cold stimulation (3℃ below the normal temperature)for 1 h,3 h,5 h and 24 h hours from 8 days age to 21 days age was measured after slaughter respectively. There is the organic specificity in expression of PCK1 gene and the expression of PCK 1 gene in liver tissue is the highest,followed by thymus gland and kidney tissue.The expression of PCK1 gene in heart has no obvious change but the expression of PCK 1 gene in liver,kidney and lung presents a rising trend.The expression of PCK1 gene in spleen thymus gland is a decline trend and the expression of PCK 1 gene in thymus gland shows a first decline and then rising trend.Cold stimulation has the up-regulated effect on expression of PCK1 gene in the liver and kidney with gluconeogenesis function.
