摘要
目的探讨Calphostin C对高转移乳腺癌MDA-MB-435S细胞侵袭能力的抑制作用。方法高转移乳腺癌MDA-MB-435S细胞系购于中国医学科学院上海细胞库,通过划痕实验、Transwell迁移实验、Transwell侵袭实验及细胞黏附实验观察Calphostin C对MDA-MB-435S细胞侵袭能力的影响。结果划痕实验结果显示,与对照组比较,实验组MDA-MB-435S细胞伤口愈合显著减慢,对照组细胞伤口24 h基本愈合,而实验组细胞伤口24 h仍未愈合。Transwell迁移和侵袭实验结果显示,实验组MDA-MB-435S细胞迁移、侵袭并穿过Transwell小室底膜的细胞数显著少于对照组(P<0.05)。细胞黏附实验结果显示,与对照组比较,实验组MDA-MB-435S细胞与纤维连接蛋白的黏附性显著降低(P<0.05)。结论 Calphostin C可显著抑制高转移乳腺癌MDA-MB-435S细胞的侵袭能力。
Objective To investigate the inhibitive effect of Calphostin C on metastasis of the breast carcinoma cells in vitro. Methods The high metastasis rate breast cancer MDA-MB-435S cells were buied from cell bank in Shanghai of Chinese Academy of Medical Sciences. The anti-metastatic effect of Calphostin C on MDA-MB-435S cells was observed by the scarification test, transwell migration test, transwell invasion test and adhesion test etc. Results The results of scarification test showed that compared with MDA-MB-435S cells in control group, the wound healing in MDA-MB-435S treated with Calphostin C in experiment group was obviously stepped down. The results of transwell migration test and invasion test revealed that the mean value of cells invaded matrigel and passed through basement membrane of chamber in experiment groups were less than those in control group. The results of adhesion test exhibited that the adhesiveness on fibronectin of MDA-MB-435S ceils in experiment group was lower than that in control group. Conclusion Calphostin C can inhibit the metastasis of the breast cancer MDA-MB-435S ceils obviously.
