摘要
通过高效毛细管电泳法(HPCE),建立一种检测齐考诺肽含量的方法。本文利用线性、精密度、稳定性、重现性、加样回收率等实验,利用HPCE法检测合成的齐考诺肽样品中齐考诺肽的含量,测定齐考诺肽在100-500mg/mL(r=0.999)范围内成良好的线性关系,精密度、重现性、稳定性、加样回收率等实验的RSD值均小于3%,加样回收率平均值为100.18%。本文首次利用HPCE法建立了检测齐考若肽含量的测定方法。结果表明此法线性好、精密度高、重现性好、结果准确,前处理简单;HPCE操作简单,分析速度快,适合齐考诺肽的含量检测。
To establish a new method for determination of ziconotide,high performance capillary electrophoresis(HPCE)was employed. HPCE was used to determine the content of ziconotide by experiments as linearity,precision,reproducibility and recoveries. The linear ranges of Ziconotide was 10-150ng/mL(r =0.9999)by HPCE,the RSD of precision,reproducibility,stability and recoveries were all less than 3%. And the average recovery by HPCE was 100.18%.A new method was established to determine Ziconotide. The method is simple,rapid,reproducible,accurate,and with good linearity,high precision. So HPCE is more suitable for content determination of Ziconotide.
