Real-time PCR分析锰对甘草β-AS基因表达的影响

摘要 目的探讨不同浓度的锰处理对甘草β-香树脂醇合成酶(β-amyrin synthase,β-AS)基因表达量的影响。方法以一年生甘草移栽苗为试验材料,设置5个锰浓度水平,分别为0,1.81,18.1,36.2和54.3 mg/L,利用Real-time PCR(荧光实时定量PCR)方法对甘草β-AS基因的Ct值进行测定并计算其相对表达量。结果随着锰处理浓度的增加,甘草β-AS基因的相对表达量呈现先升高后下降的趋势,在18.1 mg/L浓度处理时达到最大值为12.43,分别是对照(0 mg/L),36.2 mg/L和54.3 mg/L处理的2.79,1.47,2.88倍,且差异极显著(P<0.01)。结论锰对甘草β-AS基因的表达具有一定的促进作用,适当质量浓度的锰处理能够显著提高甘草β-AS基因的相对表达量。 Objective To investigate the effect of different concentrations of manganese (Mn) on the relative expression of β-AS gene in Glycyrrhiza uralensis Fiseh.Methods The transplants of one-year-old G. uralensis were subjected to five concentrations of Mn (0,1.81,18.1,36.2 and 54.3 mg/L) culturing in vermiculite. Real-time PCR was used to measure Ct vaule and work out the relative expression quantity of β-AS gene in different concentrations of Mn. Results The expression of β-AS gene of G. uralensis showed increasement with the increasement of concentration of Mn, and reached the highest 12.43 under 18.1 mg/L Mn treatment. There was very significantly difference between 18.1 mg/L concentration of Mn treatment and CK,36.2 mg/L and 54.3 nag/L( P 〈 0.01) ,and 2.79,1.47,2.88 times respectively. Conclusion Mn was provided with certain promoted function to the expression of β-AS gene, and the appropriate concentration of Mn treatment could significantly increase the relative expression quantity of β-AS gene of G. uralensis.
