摘要
目的:研究丹皮酚(Paeonol,Pae)对TNF-α诱导的人牙龈成纤维细胞(human gingival fibroblasts,HGF)黏附分子表达水平的影响。方法:使用50 ng/mL的肿瘤坏死因子(tumor necrosis factor,TNF)-α诱导刺激人牙龈成纤维细胞,后分别加入浓度为0、50、100、200 mmol/L的Pae孵育培养2、4、6天后,用Western blot法检测细胞间黏附分子(intercellular adhesion molecule,ICAM)-1以及血管细胞黏附分子(vascularcell adhesion molecule,VCAM)-1的蛋白表达水平,用逆转录多聚酶链反应(reverse transcription polymerase chain reaction,RT-PCR)检测黏附分子的mRNA表达水平。结果:TNF-α诱导的人牙龈成纤维细胞ICAM-1和VCAM-1的蛋白和mRNA表达水平均显著高于空白对照组(P<0.05);不同浓度的丹皮酚可显著降低TNF-α诱导的人牙龈成纤维细胞ICAM-1和VCAM-1的蛋白和mRNA表达(P<0.05),且随Pae浓度的升高逐渐减少。当Pae浓度达到100 mmol/L,蛋白和mRNA表达水平的抑制效果达到稳定程度(P>0.05),当Pae浓度达到200 mmol/L时,ICAM-1和VCAM-1 mRNA抑制效果达到诱导前的水平(P>0.05)。诱导时间的长短对蛋白和mRNA表达水平抑制效果没有显著影响(P>0.05)。结论:丹皮酚可显著降低TNF-α诱导的人牙龈成纤维细胞黏附分子表达水平。
Objective: To investigate the influence of Paeonol on human gingival fibroblasts (HGF) stimulated by adhesion molecules. Methods: HGF were stimulated by 50 ng/mL tumor necrosis factor (TNF)-α, then different concentrations (0, 50, 100, 200 mmol/L) of Pae were added to the experimental group. The protein and mRNA expression of intercellular adhesion molecule (ICAM)-1 and vascular cell adhesion molecule (VCAM)-1 were examined by western blot and reverse transcription polymerase chain reaction (RT-PCR). Results: The protein and mRNA expression of ICAM-1 and VCAM-1 were increased by TNF-α stimulation (P〈0.05), while Pae inhibited the expression oflCAM-1 and VCAM-1 with the increase of Pae concentration (P〈0.05). There was no significant effect of inhibition on the expression of the protein and mRNA levels in induction time (P〉0.05). Conclusion: Paeonol had the potential to provide new method for the treatment of periodontal disease.
