摘要
免疫组库是指在任何指定时间内,某个个体的循环系统中所有功能多样性B细胞或者T细胞的多态性总和.免疫组库测序技术就是通过5' RACE或者多重PCR技术将免疫球蛋白重链(IGH)或者T细胞受体beta区多态性最高的互补决定区(CDR3)捕获下来,然后进行高深度测序,研究B细胞或者T细胞的多态性,是当今一大研究亮点.免疫组库测序可以应用于医药和疫苗的研发、生物标志物的发现、微小残留病(MRD)检测、自身免疫性疾病的研究以及移植后监测等领域,因而具有重大意义.
Immune repertoire refers to the summation of the polymorphism of all functional diversity of B cells or T cells in the circulatory system of an individual at any given time. Through 5'RACE or multiplex PCR technology, immune repertoire sequencing technology can capture the complementary determining region that has the highest polymorphism in immunoglobulin heavy chain (IGH) or the beta region of T cell receptor, and then high-depth sequencing is conducted. Polymorphism of B cells or T cells is the highlight of today's stud- y. Immune repertoire sequencing can be applied to the development of vaccines and medicines, the discovery of biomarkers, the detection of minimal residual disease (MRD) , the research of autoimmune diseases,and moni- toring transplantation.
