摘要
目的人腺病毒是引起急性呼吸道感染的常见病原体,采用PCR法对人腺病毒进行分型鉴别可为人腺病毒感染的防治提供可靠依据。方法采集急性呼吸道感染患者咽拭子标本,提取DNA,采用PCR法对其进行基因分型分析。结果以500bp作为DNA分子质量标准对待测标本PCR产物进行1.2%琼脂糖凝胶电泳,46份标本中21份鉴定为腺病毒通用型(扩增片段300bp),1、2、3、4、7、21型分别有3、8、1、15、2、2份(扩增片段分别为396、393、316、305、305和507bp)。结论本院急性呼吸道感染患者感染的腺病毒以通用基因型、基因4型和基因2型为主。及时检测流行腺病毒型别及其发展规律,可为呼吸道感染类疾病的临床治疗以及防控提供指导。
Objective Human adenovirus is a common pathogen that can cause acute respiratory infection.Using PCR to type human adenovirus could provide a reliable basis for control of human adenovirus infection. Methods Throat swab samples were collected from patients with acute respiratory infections to extract DNA.Genotyping was performed using PCR. Results PCR products from test samples were subjected to electrophoresis on 1.2%agarose gel using 500 bp as a standard for the molecular weight of DNA.Twenty-one common types of adenovirus were found in 46samples(as an amplified fragment of 300bp).Adenovirus type 1was found in 3samples(as an amplified fragment of 396bp),type2 was found in 8(as an amplified fragment of 393bp),type 3was found in 1(as an amplified fragment of 316bp),type4 was found in 16(as an amplified fragment of 305bp),type 7was found in 2(as an amplified fragment of 305bp),and type 21 was found in 2(as an amplified fragment of 507bp). Conclusion Common adenovirus genotypes were found in patients with an acute respiratory infection at this hospital.These were mostly genotype 4and genotype 2.Timely detection of the types of adenoviruses that are prevalent and determining the rules governing their development could provide guidance for the clinical treatment and control of respiratory tract infections.
