电针对慢性应激抑郁模型大鼠海马内甘丙肽表达的影响 被引量:5

Influence on Expression of Galanin in Hippocampus of Electro-Acupuncture Treatment for Chronic Unpredictable Mild Stress-Induced Depression Rats

摘要 目的:观察电针对慢性应激抑郁模型(chronic unpredictable mild stress,CUMS)大鼠海马内甘丙肽(galanin,Gal)蛋白及mRNA表达的影响,探讨Gal在大鼠实验性抑郁症发病过程中的作用机制及电针干预对其的影响。方法:将36只清洁健康雄性SD大鼠,随机分为正常组、模型组及电针组,每组12只。孤养结合CUMS21d复制抑郁症模型,通过开野试验观察大鼠行为学变化,采用酶联免疫吸附法和实时定量荧光PCR检测Gal蛋白及mRNA的表达。结果:模型组大鼠开野实验水平运动及垂直运动明显减少(P<0.01),电针可逆转此变化(P<0.01);模型组大鼠海马内GAL蛋白和mRNA表达均明显下降(P<0.01),电针可改善此病理变化(P<0.05)。结论:慢性轻度不可预见性应激可使大鼠海马内GAL的表达量下降,电针可能通过增加海马内GAL表达发挥抗抑郁作用。 Objective: To investigate the influence on the expression of galanin(Gal) protein and mRNA in the hippo- campus of rat with chronic unpredictable mild stress which had the electro -acupuncture (EA) treatment and to elucidate the possible role of Gal in the development of experimental depression and the effect of EA for it. Methods : 36 adult male SD rats were randomly divided into normal control group( n = 12) ,model group ( n = 12) and EA group ( n = 12). Depression model was established by lonely raising and chronic unpredictable mild stress for 21 days. Open - field test was performed to test the behavior of rats. The expressions of hippocampus galanin and Gal mRNA were detected by using enzyme linked immunosorbent assay (ELISA) and real- time PCR techniques respectively. Results: The cross and rear score of the model group were significantly lower than that of the control group while electro - acupuncture treatment could reverse it. The GAL protein and mRNA were significantly lower in the model group while electro - acupuncture can im- prove these pathological changes. Conclusion: Chronic unpredictable mild stress can reduce GAL expressions in rats'hip- pocampus and electro - acupuncture treatment can increase the expression of GAL to prevent depression.
