摘要
目的:研究Pax1在乳癌组织中的表达及其对乳癌MDA-MB-231细胞生物学行为的影响。方法:采用Western blot检测200乳腺原发癌和癌旁组织中Pax1蛋白的表达。采用化学合成针对Pax1基因的两个小干扰RNA(siRNA-Pax1-1和siRNA-Pax1-2)下调该基因的表达,应用MTT和Transwell实验检测其对MDA-MB-231细胞生物学行为的影响。结果:Pax1在乳腺原发癌组织中的表达低于相应的癌旁组织,沉默Pax1表达可促进乳癌细胞增殖、迁移和侵袭(F=52.413和72.741,P<0.001)。结论:Pax1有望成为乳癌早期诊断和预测预后的生物分子标志物。
Aim: To investigate the expression of paired boxed gene 1( Pax1) gene in breast cancer tissue and to explore the influence on proliferation status,migration ability and metastatic ability of breast cancer cells MDA-MB-231.Methods: Western blot were used to examine the expression of Pax1 in samps,including 200 normal tissues and 200 primary tumors. MTT and transwell were used to analyze the ability of proliferation,migration,and invasion in Pax1 siRNAtransfected MDA-MB-231 cells. Results: The expression of Pax1 was lower in breast cancer tissues than other tissues.Furthermore,knockdown of Pax1 expression resulted in increased proliferation and invasion in MDA-MB-231 cells in vitro( F =52.413 and 72.741,P〈0. 001). Conclusion: The expression of Pax1 gene shows and obvious downgraded in breast cancer tissue. Low expression of Pax1 gene can increase apeptosis and proliteration of MDA-MB-231 cells,which may provide a strategy for blocking breast cancer and progression.
