Content Comparison of Abrusamide A in Abrus Herb Leaves from Different Varieties

目的 建立鸡骨草叶中鸡骨草甲素含量测定方法,并对其不同来源和品种进行比较分析。方法采用高效液相色谱法,色谱条件为：Agilent ZORBAX SB-C18色谱柱（250mm×4.6mm,5μm）,以甲醇（A）-0.2%乙酸水（B）为流动相采用梯度洗脱,流速为1.0 mL·min-1,检测波长275nm。结果 鸡骨草甲素在0.429~8.58μg范围内线性关系良好（r=1.0000）,平均加样回收率为98.92%,RSD=2.8%（n=6）,不同来源和品种鸡骨草叶中鸡骨草甲素含量存在明显差异。结论 该方法简便、准确、分离度和重现性好,可用于鸡骨草叶中鸡骨草甲素的含量测定。

Objective To establish a method to determine and analyse the content of abrusamide A in theleaves ofAbrus cantoniensis （AC） and A. mollis （AM） from different sources and varieties. Methods An HPLCmethod was carried out with an Agilent ZORBAX SB-C18 （250 mm×4.6mm, 5μm） column. The mobile phaseswere methanol （A） and 0.2% acetic acid solution（B） with gradient condition. The flow rate was 1.0 mL·min^-1 andthe detection wavelength was set at 275 nm. Results The linear range of abrusamide A was 0.429-8.58pg. Theaverage recovery was 98.92% with RSD of 2.8%（n=6）. The content of abrusamide A was markly different amongdifferent sources and varieties of Abrus Herb leaves. Conclusion The method was simple, accurate, reliable,reproducible and suitable for the content determination of abrusamide A in the leaves of AC and AM.
