摘要
目的建立药用植物多糖离子色谱分析的在线酶水解方法。方法以苦瓜多糖为例,将苦瓜粗多糖通过特异性酶水解成小分子单糖,小分子单糖透过渗析膜到达渗析液中,大分子化合物截留在膜外,达到消除基质的目的。然后用蠕动泵连接色谱六通阀在线抽取样品进入离子色谱分析。结果苦瓜多糖在线水解1 h左右时已水解完全,各组分峰分离效果良好。结论建立了一种用于植物多糖在线评价的模式及研究技术方法,为植物多糖的分析方法和推广应用提供实验基础。
Objective To establish a method for on-line enzyme hydrolysis of medicinal plantpolysaccharides.Methods Taking momordica polysaccharide as an example, macromolecular polysaccharidematerial was hydrolyzed into small molecular sugars with specific enzyme, and reached the dialysis fluid throughthe dialysis membrane.Monosaccharide components were determined with ion chromatography analysis by on-linesampling.Results The main contains of momordica polysaceharide were separated well after on-line hydrolysisabout 1 hour.Conclusion An on-line evaluation model and method was established for plant polysaccharides. Itprovides an experimental basis for the analysis and application of plant polysaccharides.
