摘要 目的:探讨重组人白细胞介素24(rhIL-24)对人肺腺癌耐顺铂细胞株A549/DDP细胞的体外抑制效应;探讨rhIL-24联合顺铂体外诱导A549/DDP细胞凋亡、细胞周期的影响。方法:rhIL-24、顺铂(DDP)及rhIL-24+DDP干预A549/DDP细胞,用CCK-8法检测A549/DDP细胞增殖抑制率;用流式细胞仪检测细胞凋亡及细胞周期的变化。结果:rhIL-24联合DDP作用24 h对人肺腺癌A549/DDP细胞增殖抑制率分别为25.2%,与单独用DDP组比较P<0.05有显著性差异;流式细胞仪检测rhIL-24浓度为160 ng/ml作用24 h对人肺腺癌A549/DDP细胞凋亡率为19.0%;细胞周期检测中rhIL-24作用A549/DDP细胞24 h后G2期增多。结论:rhIL-24能抑制A549/DDP细胞增殖,对正常细胞无毒性,联合DDP组效果优于单独用药组,具有化疗增敏效应;rhIL-24能诱导A549/DDP细胞凋亡和影响细胞周期。 Objective: To investigate the inhibitory effects of recombinant human interleukin-24 (rhIL-24) on human lung ade- nocarcinoma cell lines A549 in vitro, and to observe the changes in apoptosis and cell cycle. Methods: Cell proliferation was detected by CCK8 assay, and the changes in apoptosis and cell cycle of A549/DDP were detected by flow cytometry after the treatment with rhIL- 24 alone, DDP alone and rhIL-24 combined with DDP. Results: The treatment with rhIL-24 significantly inhibited the growth of human lung adenocarcinoma cell lines A549/DDP in vitro, rhIL-24 ( 160 ng/ml) combined with DDP after 24 h, resulted in a 25.2% growth inhibition,which showed a significant difference with DDP alone group. The apoptosis rate of A549/DDP detected by flow cytometry in combination group was 19% Furthermore,cell cycle detection indicated that the percentage of A549/DDP in G2 phases increased re- spectively in rhIL-24 alone group. Conclusion: The treatment with rhIL-24 led to the growth inhibition of A549/DDP, non-toxic to normal cells ,rhlL-24 combine with DDP ring about a striking effect than rhIL-24 single work. rhlL-24can enhances the effect of chemo- therapy, rhIL-24 can induce apoptosis and cell-cycle alteration of human lung adenocarcinoma A549/DDP.
