甘氨双唑钠通过影响喉癌细胞的细胞周期实现放射增敏作用 被引量:1

摘要 目的探讨甘氨双唑钠对放射线照射后喉癌细胞各细胞周期细胞比例的影响。方法选择喉癌Hep-2细胞,共分为5组。对照组:癌细胞不做任何处理;放射组:癌细胞仅接受6 MV-X线照射,剂量率300 cGy/min,剂量5 Gy;甘氨双唑钠组:予0.8 mmol/L甘氨双唑钠100μl作用4 h;甘氨双唑钠+放射组:同甘氨双唑钠组及放射组;AT突变基因(AT mutated,ATM)阻断+甘氨双唑钠+放射组:KU55933+0.8 mmol/L甘氨双唑钠的培养液培养4 h后行5 Gy照射。分别采用流式细胞术和蛋白免疫印迹杂交技术检测细胞周期的变化及Bax蛋白表达量。结果经甘氨双唑钠作用和5 Gy X线照射后,喉癌细胞G0/G1期细胞明显降低,Bax蛋白表达水平明显升高,与不做任何处理、单独X线照射、单独甘氨双唑钠作用及ATM阻断+甘氨双唑钠+放射治疗的喉癌细胞比较差异均有统计学意义(P<0.05)。结论甘氨双唑钠可通过影响喉癌细胞的细胞周期实现放射增敏作用。 Objective To evaluate the cell cycle of laryngeal cancer cells after the intervention of combined sodium glycididazole and radiation. Methods Laryngeal cancer Hep-2 cells were divided into five groups:the control group with no treatment, the irradiation group (6MV-X radiation, 300 cGy/min, 5 Gy), the sodium glycididazole group ( treated with 100 μl of sodium glycididazole at the concentration of 0. 8 mmol/L, 4 h), and the sodium glycididazole plus irradiation group (treated with the same above) and the KU55933 plus sodium glycididazole (0. 08 mmol/L, 4 h) and irradiation (5 Gy) group. The cell cycle and Bax protein expression of laryngeal cancer cells were detected by FCM and Western blotting, respec-tively. Results In sodium glycididazole plus irradiation group, the cell proportion of G0/G1 phase was declined significantly and the Bax protein expression of laryngeal cancer cells increased, and the differences were significant when compared to other groups (P〈0. 05). Conclusion Sodium glycididazole enhances radiosensitivity of laryngeal cancer cells through regulating the cell cycle.
