ManLAM对B细胞表达抑制性细胞因子IL-10的影响

摘要 目的:研究结核分枝杆菌细胞壁上的甘露糖修饰的脂阿拉伯甘露聚糖( mannose-capped lipoarabinomanan,ManLAM)对小鼠B细胞的影响。方法用结核分枝杆菌标准菌株H37Rv来源的ManLAM刺激小鼠脾细胞或者磁珠纯化的B细胞12 h,然后用流式细胞仪检测B细胞对抑制性细胞因子IL-10的表达情况。结果 iH37Rv(灭活的H37Rv)和ManLAM刺激小鼠脾脏细胞后,能促进其中的B细胞表达IL-10,在8-12 h内表达量达到高峰,且ManLAM能直接作用于小鼠B细胞,促进其对IL-10的表达。结论结核分枝杆菌可以通过其细胞壁上的ManLAM促进B细胞表达抑制性细胞因子IL-10,抑制机体免疫系统的正常功能,从而逃避宿主的杀害。 Objective To study the effects of ManLAM sourcing the cell wall of Mycobacterium tuberculosis on B cell function of mouse. Methods We stimulated mice spleen cells and B cells of magnetic beads purification by the ManLAM sourced in Standard strain H37Rv mycobacterium tuberculosis for 12 hours. And then we detected B cells' expression of inhibitory cytokine IL-10 by flow cy-tometry instrument. Results After mice spleen cell had been stimulated by iH37Rv ( inactivated H37Rv) and ManLAM,its B-cells can be promoted to express IL-10. Within 8 to 12 hours,the expression quality would peak,and ManLAM could act directly on B cells in mice,and promoted its expression of IL-10. Conclusions Mycobacterium tuberculosis can promote B cells to express inhibitory cyto-kines IL-10 by the ManLAM on its cell wall and inhibit the normal function of the body's immune system,so as to avoid the host's killing.
