摘要
目的:建立高效液相色谱法测定大鼠血浆中黄芩苷和盐酸小檗碱,进行药代动力学研究。方法:以紫草酸为内标,采用Agilent C18色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.3%磷酸三乙胺(22∶78),流速1.0 mL·min-1,检测波长278 nm,进样体积20μL。结果:黄芩苷和盐酸小檗碱呈良好线性关系;回收率分别为82.1%~87.1%、80.1%~87.5%,日内、日间精密度的RSD均小于10%。结论:为黄芩苷、盐酸小檗碱的药代动力学研究提供方法学依据。
Abstract: Objective: To develop an HPLC method for the determination of baicalin and berberine hydrochloride in rat plasma for the subsequent pharmacokinetic study. Methods : Purple oxalic acid as internal standard, Agilent C18 Chromatographic column (4.6 mm× 250 mm, 5 μ m )with the column temperature of 30 ℃ was used. The mobile phase consisted of three-0.3% phosphate triethylamine ( 22: 78 ) with flow rate of 1.0 mL · min-i, and the detection wavelength was set at 278 nm and the injection volume was 20 μ L. Results : The developed method had a good linearity in the ranges. Low and high recoveries of the 2 mass concentration were 82.1% - 87.1%, 80.1% - 87.5%. Days, inter-day precision of RSD was less than 10%. Conclusion : Pharmacokinetic studies provide methodological basis for baicalin and berberine hydrochloride.
