十全大补汤联合5-氟尿嘧啶对移植大肠癌小鼠免疫抑瘤作用影响 被引量:1

Effect of Mice Colorectal Cancer's Immune Inhibitory of Shiquan Dabu Decoction Combined with 5-Fluorouracil

摘要 目的:研究中医药经典方剂十全大补汤(以下简称sqdbt)联合临床常用化疗药物5-氟尿嘧啶(以下简称5-fu)对HT29移植性大肠癌小鼠的抑瘤作用及对小鼠免疫功能的影响。方法:以HT29移植性大肠癌小鼠为研究对象,随机分成单纯肿瘤组、sqdbt组、5-fu组、sqdbt+5-fu组,计算胸腺指数,TUNEL法检测肿瘤细胞凋亡,ELISA法检测血清中TNF-α浓度。结果:治疗组胸腺指数均大于单纯肿瘤组;TUNEL法检测凋亡细胞sqdbt+5-fu组最高为11.35;血清中TNF-α浓度sqdbt+5-fu组最高,可达44.88 pg/mL。结论:十全大补汤联合5-氟尿嘧啶在抑制大肠癌小鼠肿瘤细胞生长和减少化疗药物对机体免疫损伤方面,作用优于单独使用化疗药物。 Objective: To study the effect of TCM classical prescription of Shiquan Dabu Decoction ( hereinafter referred to as sqdbt ) combined with chemotherapy drug 5- fluorouracil ( 5-FU ) on the immune function of the mice and the anti-tumor effect of HT29 transplantation of eoloreetal cancer in mice. Methods: HT29 mice transplanted colorectal cancer were as the research object and randomly divided into simple tumor group, sqdbt group, 5-FU group, sqdbt+5-fu group. The thymus index was measured. TUNEL method was used to detect the apoptosis of tumor cells, and ELISA to test TNF- α in the serum. Results : The thymus index of treatment group was higher than that of simple tumor group. The apoptotic cells in sqdbt+5- fu group tested by TUNEL was up to 11.35. The serum concentration of TNF- α in sqdbt+5-fu group was the highest, up to 44.88 pg/mL Conclusion : Shiquan Dabu Decoction combined with 5- fluorouracil has better effect on the inhibition of colorectal tumor cells growth and reducing the body immune damage due to chemotherapy drugs than the use of chemotherapy alone.
