摘要
目的:采用高脂高糖饲料喂养和单侧肾脏切除后进行腹腔注射链脲佐菌素(STZ)的方法进行比较,探讨不同方法建立糖尿病肾病大鼠的最佳方案。方法:清洁级Wister大鼠30只,随机分为3组,每组10只,A组为正常对照组,B组为高脂高糖+STZ组(35 mg/kg腹腔注射),C组为单侧肾切除+STZ组(35 mg/kg腹腔注射),造模后2周、6周、10周测定体重、血糖,收集24 h尿液测定尿微量白蛋白,实验结束时取肾脏病理切片观察。结果:高脂高糖+STZ组和单侧肾切除+STZ组的血糖、尿微量白蛋白与正常对照组比较差异显著,高脂高糖+STZ组成模率高,单侧肾切除+STZ组模型稳定。结论:单侧肾切除+STZ组(35 mg/kg腹腔注射)可以作为建立糖尿病肾病大鼠模型的理想方案。
Objective: To make comparison by the way of HFSD feed and intraperitoneal injection with streptozotocin after unilateral nephrectomy, and then to discuss the ideal plan among the different methods of diabetic nephropathy ( DN ) rat model building. Methods : Thirty clean-grade Wister rats were randomly divided into three groups with 10 rats in each: normal control group A, group B with high fat and high glucose+STZ( intraperitoneal injection of 35mg/mL ), and group C with Uninephrectomy +STZ( intraperitoneal injection of 35mg/mL ) .Two weeks, six weeks and ten weeks after model building, we measured the weight and blood glucose respectively, collected 24-hour urine to measure microalbuminuria and took the renal pathological section for observation when the experiment was finished. Results : There was a great difference in the blood glucose and mieroalbuminuria between the group with high fat and high glucose+STZ and group Uninephrectomy+STZ and the normal control. Group with high fat and high glueose+STZ had a higher incidence while group with Uninephrectomy+STZ was more stable in modeling. Conclusion: Group with Uninephrectomy +STZ ( intraperitoneal injection of 35mg/mL ) can he taken as the ideal plan to build the DN rat models.
