护肾复方对2型糖尿病大鼠肾脏保护作用研究 被引量:1

摘要 目的研究护肾复方对糖尿病大鼠肾脏的保护作用及对大鼠肾脏PAI-1的影响。方法雄性Wistar大鼠,通过高脂饮食喂养加小剂量链脲佐菌素建立2型糖尿病大鼠模型,造模成功后随机分为模型组、护肾复方高剂量组和护肾复方低剂量组,并设正常对照组,每组8只。用药持续治疗12周,并定期检测血糖,于治疗第12周末检测肌酐、24小时尿蛋白及计算内生肌酐清除率,最后取肾脏称重并计算肾体重比,HE染色光镜下观察肾组织结构的变化,通过免疫组织化学定位并定量PAI-1的表达。结果与正常组相比,模型组大鼠血糖显著升高,肾功能损害,PAI-1表达增多,肾脏病理学观察显示肾小球肥大,基底膜增厚,系膜细胞增生,系膜基质增多,肾小管间质胶原纤维明显增加。护肾复方能显著降低糖尿病大鼠空腹血糖,改善肾功能,下调PAI-1在肾组织的表达,明显改善肾脏病变。结论护肾复方降低糖尿病大鼠血糖,改善肾功能及降低PAI-1在肾组织的表达,具有一定的肾脏保护作用。 Objective To investigate the effect of Hushen decoction on renal function of diabetic rats. Method Male Wistar rats were induced by high - fat - high - sucrose feeding and injection of low dose streptozotocin to establish type 2 diabetic model rats. Then, the rats were randomly divided into normal group, model group, high - dosage Hushen decoction group and low - dosage Hushen decoction group. We detect fasting blood glucose periodically and kidney function index at the end of the test, finally take the kidney and its weight and the kidney index were calculated. Using HE staining to observe the structural changes of renal tissue. Using immunohistochemical to observe the location of PAI - 1 expressed. Result Hushen decoction can significantly reduce fasting blood glucose, improve renal function index, inhibit the expression of PAI- 1 in the kidney tissues and reduce renal parenchyma pathological damage by high blood glucose in diabetic rats. Conclusion Hushen decoction can lower blood sugar levels and suppress the expression of PAI- 1 in the kidney, and may have certain protective effects on renal function in diabetic rats.
