摘要
目的:探讨玛咖多糖对黑腹果蝇寿命和抗氧化作用的影响。方法:采用黑腹果蝇为研究对象,收集8 h内羽化未交配的果蝇,雌雄分开,将果蝇随机分为对照组,玛咖多糖低、中、高剂量组(质量浓度分别为0.03,0.08,0.24 g·L-1)。每组10管,每管20只,每2 d记录果蝇的死亡数,每4 d更换1次培养基,直至果蝇全部死亡。每组全部果蝇死亡天数的平均数为该组的平均寿命,每组中半数果蝇死亡的天数为该组的半数死亡时间,每组中最后死亡的20只果蝇存活天数的平均数为该组的最高寿命。测定百草枯和过氧化氢急性氧化损伤下的存活时间、超氧化物歧化酶(SOD)活性、过氧化氢酶(CAT)活性以及脂褐素含量。结果:与对照组相比,玛咖多糖高剂量组能延长果蝇平均寿命及最高寿命(P<0.05或P<0.01),中、高剂量组能延长过氧化氢和百草枯急性氧化损伤下果蝇的存活时间(P<0.05或P<0.01),各剂量组均能提高SOD和CAT活性,降低脂褐素含量(P<0.05或P<0.01)。结论:玛咖多糖可以提高果蝇的抗氧化能力,抑制脂质过氧化,延长果蝇寿命。
Objective:To investigate the effect of Maca polysaccharide on the life span and antioxidative ability of Drosophila melanogaster.Method:Eight hour eclosion unmated D.melanegaster were collected.Male and female were separated,D.melanogaster were randomly divided into 4 groups:control group,Maca polysaccharide group (0.03,0.08,0.24 g · L-1).The life span,the survival time in paraquat and hydrogen peroxide treatment,the activities of superoxide dismutase (SOD),catalase (CAT) and the content of lipofuscin were determined.Result:Compared with the control group,the high dose group extends the maximum life span and mean life span of D.melanogaster (P < 0.05 or P < 0.01),exhibiting a dosage effect.The middle dose group and the high dose group can improve the survival time in paraquat and hydrogen peroxide treatment.The activities of SOD and CAT increase,meanwhile the content of lipofuscin decreases in the experiment.Conclusion:Maca polysaccharide might have a satisfactory effect on life span and anti-oxidant.
